چکیده

Methods Nineteen adult sheep were anesthetized, mechanically ventilated and invasively monitored. Renal blood-flow was measured continuously through a left lumbotomy using a peri-vascular flow probe (Transonic, USA) and an aortic clamp was positioned above the renal arteries. After surgical preparation and stabilization, animals were randomized into 3 groups (SHAM=5, I-R=7, I-R+BH4=7). SHAM animals underwent surgical preparation but no aortic clamping was performed. The I-R groups were exposed to 1 hour of aortic ischemia. The I-R+BH4 group received 20 mg/kg of BH4 before aortic clamping. Animals were followed for a maximum of 6 hours after reperfusion. Renal microcirculation was evaluated at baseline, and 1, 4 and 6 hours after reperfusion using Sidestream Dark Field video-microscopy (Microvision Medical, Netherlands). We calculated perfused small vessel density (PVD), proportion of perfused small vessels (PPV) and heterogeneity of PPV (PPV-HI). Data were analyzed using the generalized

Treatment With Tetrahydrobiopterin Overcomes Brain Death–Associated Injury in a Murine Model of Pancreas Transplantation

Brain death (BD) has been associated with an immunological priming of donor organs and is thought to exacerbate ischemia reperfusion injury (IRI). Recently, we showed that the essential nitric oxide synthase co-factor tetrahydrobiopterin (BH4) abrogates IRI following experimental pancreas transplantation. We therefore studied the effects of BD in a murine model of syngeneic pancreas transplanta...

Beneficial effect of tetrahydrobiopterin on the survival of rats exposed to hepatic ischemia-reperfusion injury.

INTRODUCTION The protective role of nitric oxide (NO) against hepatic ischemia-reperfusion (I/R) injury remains controversial. In this study we investigated the effect of tetrahydrobiopterin (BH4) on the survival of rats exposed to an hepatic I/R injury. METHODS The rats were subjected to 100 minutes of 70% hepatic ischemia 30 minutes after administration of BH4 or saline. A specific inducibl...

Cardioprotective effects of tetrahydrobiopterin in cold heart preservation after cardiac arrest.

BACKGROUND It has recently been shown that tetrahydrobiopterin (BH4), an essential cofactor of nitric oxide synthase (NOS), reduces ischemia-reperfusion myocardial injury. The aim of this study was to determine if supplementation with BH4 after cardiac arrest followed by cold heart preservation would exert a cardioprotective effect against ischemia-reperfusion injury. MATERIALS AND METHODS Is...

Effects of tetrahydrobiopterin on endothelial dysfunction in rats with ischemic acute renal failure.

The role of nitric oxide (NO) in ischemic renal injury is still controversial. NO release was measured in rat kidneys subjected to ischemia and reperfusion to determine whether (6R)-5,6,7,8-tetrahydro-L-biopterin (BH4), a cofactor of NO synthase (NOS), reduces ischemic injury. Twenty-four hours after bilateral renal arterial clamp for 45 min, acetylcholine-induced vasorelaxation and NO release ...

Candesartan Attenuates Ischemic Brain Edema and Protects the Blood–Brain Barrier Integrity from Ischemia/Reperfusion Injury in Rats

Background: Angiotensin II (Ang II) has an important role on cerebral microcirculation however, its direct roles in terms of ischemic brain edema need to be clarified. This study evaluated the role of central Ang II by using candesartan, as an AT1 receptor blocker, in the brain edema formation and blood-brain barrier (BBB) disruption caused by ischemia/reperfusion (I/R) injuries in rat. Methods...
